Rats are used to make rat monoclonal anti mouse antibodies. There are less rat- than mouse clones however. Rats genes from rodents of the genus Rattus norvegicus are often studied in vivo as a model of human genes in Sprague-Dawley or Wistar rats.
Reverse transcription primers are used in PCR but in vivo reverse transcription begins when the viral particle that enters the cytoplasm of a target cell with its reverse transcriptase. The viral RNA genome enters the cytoplasm as part of a nucleoprotein complex that has not been well characterized. The process of reverse transcription generates, in the cytoplasm, a linear DNA via an intricate series of steps. This DNA is collinear with its RNA template, but it contains terminal duplications known as the long terminal repeats (LTRs) that are not present in viral RNA . Extant models for reverse transcription propose that two specialized template switches known as strand-transfer reactions or “jumps” are required to generate the LTRs.
The test principle applied in this kit is Sandwich enzyme immunoassay. The microtiter plate provided in this kit has been pre-coated with an antibody specific to Telomerase Reverse Transcriptase (TERT). Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody specific to Telomerase Reverse Transcriptase (TERT). Next, Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. After TMB substrate solution is added, only those wells that contain Telomerase Reverse Transcriptase (TERT), biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450nm ± 10nm. The concentration of Telomerase Reverse Transcriptase (TERT) in the samples is then determined by comparing the O.D. of the samples to the standard curve.
